I gather you still get that message, even with the CD in the drive?

One possibility is that the drive letter has changed. This can happen
if you've added or removed some hardware. The CD drive letter is stored
in 'startup.mar', as Konstantin mentioned. Edit it in Notepad or Word
or similar editor, and look for these lines:

63 real CD Z
18 drive Z:\

In place of 'Z', you'll see the (wrong) CD drive letter. Correct it,
save, and Guide ought to run Just Fine.
